非小细胞肺癌(NSCLC)目前仍是全球的主要健康威胁之一。尽管目前对于NSCLC有几种有效的治疗方法(如手术、放疗、化疗和靶向治疗),但NSCLC患者的预后仍不尽人意。在过去的几年里,免疫疗法已成为一种新型的癌症治疗方法。

在癌细胞形成临床上可识别的肿瘤之前,免疫系统对于其消除具有重要作用。为抵抗这种免疫压力,一些癌细胞通过与调节性T细胞上表达的免疫检查点分子进行相互作用并逃脱免疫系统的监视,称为免疫逃逸。免疫逃逸是肿瘤发生的重要机制。

SKIL在NSCLC组织中的表达状态

该研究旨在探究SKIL在NSCLC的肿瘤发生和免疫逃逸中的作用。


研究人员通过定量PCR、蛋白质印迹实验以及免疫组化等方法,分析NSCLC细胞系、临床样品和邻近正常组织中的SKIL的表达水平。并通过集落形成、transwell小室实验、MTT分析以及移植瘤小鼠模型评估了NSCLC细胞的恶性表型。

沉默TAZ通过激活STING信号通路促进SKIL过表达肿瘤的T细胞浸润

研究人员发现,相比于邻近正常组织,NSCLC组织中的SKIL的表达水平更高。沉默SKIL可以抑制NSCLC细胞的恶性程度并促进T细胞浸润作用。敲低SKIL的表达可以通过下调TAZ来抑制NSCLC细胞的自噬作用并激活STING信号通路。而沉默TAZ则能够消除过表达SKIL对NSCLC细胞恶性程度以及自噬作用的影响。进一步研究显示,对细胞自噬作用的抑制能够逆转SKIL/TAZ过表达对STING信号通路的影响。


总而言之,SKIL能够通过上调TAZ的表达并促进细胞自噬以及抑制下游STING信号通路,促进NSCLC的肿瘤发生和免疫逃逸。
