     银屑病在皮肤上引起红色、厚厚的鳞片状斑块,影响全球人口的2-3%。角质层增厚是由于表皮中的主要细胞类型角质形成细胞的异常增殖所致。银屑病的常规治疗包括全身应用可能导致肝和肾毒性的免疫抑制剂(如甲氨蝶呤),或局部应用皮质类固醇(如倍他米松)。目前许多药物的疗效和安全性都很差,因为它们存在非特异性给药和表皮滞留不良的问题。要治疗银屑病,药物必须穿过增厚的疏水角质层屏障并进入表皮细胞。

    纳米粒(NPs)有望用于银屑病的局部给药,因为它们可以被设计成改善药物的溶解性、稳定性、渗透性和皮肤保持性。脂质体和脂质纳米粒疏水性有利于皮肤渗透,是输送抗银屑病药物(如维生素D类似物、13种核酸、14种−16和多肽)的主流载体。

     金纳米颗粒(AuNPs)是脂基纳米颗粒的有前途的替代载体,因为它们可以通过TEM18和无染料共聚焦反射显微镜在细胞中可视化,并且它们的细胞摄取可以通过电感耦合等离子体质谱(icp-MS)来定量;金纳米颗粒的这种吸引人的特性使得人们能够全面地研究它们的“皮肤−纳米”相互作用。局部给药的一个关键挑战是它们需要辅料来穿透银屑病的鳞片,但是。在评估其疗效之前,很少有研究确凿地证明了将载药的AuNPs输送到表皮细胞。选择AuNPs作为抗银屑病纳米药物的另一个原因来自于它们用于治疗类风湿性关节炎、癌症和其他疾病的固有抗炎和抗血管生成特性。

     这种亚15 nm纳米粒子不含已知的化学或生物抗银屑病药物,它包含一个3 nm的金核和一个由1000 Da聚乙二醇链组成的外壳,由30%的十八烷基链修饰。当应用于无辅料的咪喹莫特诱导的银屑病小鼠时,纳米颗粒可以穿过角质层,优先进入角质形成细胞。将纳米颗粒与咪喹莫特同时应用可预防银屑病,并下调白细胞介素-17信号通路下游富含的与表皮过度增殖和炎症相关的基因。在牛皮癣确诊后应用纳米颗粒治疗牛皮癣,与标准的类固醇和维生素D类似物疗法一样有效,但不会脱发和皮肤起皱。纳米颗粒不会在主要器官中积聚,也不会产生长期毒性。

       首先,已经证明了纳米粒的大小和烷基负载量是将纳米粒局部输送到健康和银屑病皮肤表皮细胞的关键参数。含有官能团的纳米颗粒的表面工程(在这种情况下是通过烷基化)可以克服表皮角质形成细胞的主要传递障碍,用于治疗银屑病。最后,已经证明AU3@PEGoctadecyl30%纳米粒治疗银屑病与含有类固醇和维生素D类似物的药物一样有效,但没有在主要内脏的长期滞留或长期毒性。而且它代表了一种简单、安全和有效的银屑病治疗方法。
