草酸盐(Ox)是一种由肾脏产生的代谢性终产物,与多种病理状态有关。草酸盐在体内的积累是导致草酸钙肾结石的因素之一。为了模拟体内高浓度的Ox环境,我们使用肾小管上皮(HK-2)细胞建立了高Ox的体外模型。评估细胞活力和增殖以评估Ox的不同浓度(0,0.25,0.5,1,2,4,5,8和10mM)对HK-2细胞的作用以选择提取外泌体的最佳浓度和时间。结果显示,使用0,1或2mM Ox处理可改变外泌体的

结果显示,使用0,1或2mM Ox处理可改变外泌体的形态和分泌能力。随着Ox浓度的增加,外泌体的峰值和平均粒径减小,但外泌体颗粒浓度、外泌体RNA和外泌体蛋白质的水平增加。不同的胞外囊泡亚型中,外泌体的大小、分布和分泌速率以及RNA和蛋白质含量是不同的。此外,三种亚型的外泌体在微环境中传递不同的信号因子。

因此,我们推测三种亚型的外泌体在细胞间信号传递和CaOx肾结石形成中可能发挥不同的作用。
