环状GMP-AMP合成酶(cGAS)是细胞质DNA的重要传感器,对先天性免疫反应和自身免疫具有重要的介导作用。调节cGAS的活性和稳定性为治疗病毒性或自身免疫性疾病提供了潜在的策略。

最近,研究人员发现,泛素特异性蛋白酶29(USP29)可以脱泛素化并稳定cGAS,促进细胞抗病毒反应和自身免疫。USP29的敲低或敲除严重损害了单纯疱疹病毒1(HSV-1)-或细胞内DNA诱导的I型干扰素(IFNs)和促炎症细胞因子的表达。

一致的是,与野生型小鼠相比,Usp29m/m小鼠在HSV-1感染后产生I型IFNs和促炎性细胞因子减少,对HSV-1感染的敏感度较高。

此外,在Trex1-/-小鼠中,USP29的基因敲除消除了可检测到的病理和分子自身免疫表型。

从机理上讲,USP29与cGAS相互作用,使cGAS与K48连接的多泛素链脱离,并在未感染细胞中或HSV-1感染后稳定cGAS。将cGAS重组到Usp29-/-细胞中,可以完全拯救I型IFN的诱导和HSV-1感染后的细胞抗病毒反应。

因此,该研究结果揭示了USP29在对抗DNA病毒和自身免疫性疾病的先天性抗病毒反应中的关键作用,并为cGAS的调控提供了启示。
