最近,有研究人员在2个意大利前列腺癌群体(n=157和182)和2个美国前列腺癌群体(n=214和298)中进行了案例-对照研究。研究人员在NTAT群体中利用线性回归评估了选择的3个DNMT3B变异与全部DNA甲基化之间的相关性;在所有群体中利用Cox比例风险回归评估了3个DNMT3B变异与PCa死亡率之间的相关性;还利用逻辑回归评估了DNMT3B的表达与致死性PCa之间的相关性,并与公共数据库进行了重复性比较。研究发现,在肿瘤组织中,rs1569686的TT基因型与LINE-1低甲基化相关(β=2.71, 95% CI: 5.41, 0.05),而DNMT3B与PCa死亡率之间没有发现相关证据证明其相关性。在2个美国群体中,DNMT3B的表达与致死性PCa一致性相关(3rd vs 1st tertil,联合群体:OR=2.04, 95% CI: 1.13, 3.76);另外,上述相关性在TCGA和MSKCC数据中能够得到重复(3rd vs 1st tertile, TCGA: HR=3.00, 95% CI: 1.78, 5.06; MSKCC: HR=2.22, 95% CI: 1.01, 4.86)。

最后,研究人员指出,尽管DNMT3B变异和PCa死亡率之间没有证据证明其相关性,在肿瘤组织中,rs1569686的TT基因型与LINE-1的低甲基化相关,并且DNMT3B的mRNA表达与致死性PCa风险的增加相关。
