神经发生是神经干细胞增殖分化产生新生神经元的过程,对哺乳动物大脑的正确发育及功能连接建成至关重要。在胚胎发育过程中,室管膜区域的神经上皮细胞通过对称分裂扩增祖细胞库,当神经上皮增厚至假复层室壁时,神经上皮细胞转化为放射状胶质细胞,即胚胎神经干细胞(eNSCs),后者直接产生神经元,或经中间前体细胞间接产生神经元。与此同时,一部分的 eNSCs 开始缓慢增殖并存留下来成为侧脑室室管膜亚区和海马齿状回

神经发生是神经干细胞增殖分化产生新生神经元的过程,对哺乳动物大脑的正确发育及功能连接建成至关重要。在胚胎发育过程中,室管膜区域的神经上皮细胞通过对称分裂扩增祖细胞库,当神经上皮增厚至假复层室壁时,神经上皮细胞转化为放射状胶质细胞,即胚胎神经干细胞(eNSCs),后者直接产生神经元,或经中间前体细胞间接产生神经元。与此同时,一部分的 eNSCs 开始缓慢增殖并存留下来成为侧脑室室管膜亚区和海马齿状回颗粒下区的成体神经干细胞(aNSCs)。尽管成体神经发生的增殖分化过程与胚胎期神经发生相类似,但其在增殖速率、分化速度及细胞微环境变化等方面仍然存在较大差异。这两个阶段神经发生的差异调节的内在机制仍然不清楚。

中国科学院遗传与发育生物学研究所郭伟翔研究组的研究结果表明,在神经干细胞中特异性敲除 RNA 结合蛋白 HuR,导致成年神经发生缺陷,对胚胎期神经发生没有影响。在 eNSCs 中,HuR 主要定位于细胞质中,随着发育的进行,aNSCs 中 HuR 主要定位于细胞核中。进一步分子机制分析结果表明,HuR 调控黏着斑激酶(FAK)的选择性剪切。在 HuR 敲除的 aNSCs 中会形成 5’-UTR 变短的 mRNA,导致 FAK mRNA 翻译增加,FAK 信号过度激活。在 HuR 条件性敲除小鼠中药物抑制 FAK 的活性能改善成年神经发生缺陷及海马依赖性的学习和记忆障碍。这一工作提出在发育过程中,HuR 在 eNSCs 和 aNSCs 的细胞质和细胞核中易位表达,可能是胚胎神经发生和成年神经发生差异调节的分子机制之一。

该成果于 2019 年 12 月 3 日在线发表于 Cell Reports 上(DOI:10.1016/j.celrep.2019.10.127)。郭伟翔组博士研究生王玉晗,博士后郭野及已毕业博士生汤常永为该论文第一作者,郭伟翔研究员为通讯作者。该研究得到了中科院遗传发育所曹晓风研究组在生物信息学分析上的帮助,以及东北师范大学朱筱娟教授研究组在实验上帮助。该研究得到了国家自然科学基金委和北京市科委的资助。
