循环肿瘤细胞(CTC)被认为是监测转移性乳腺癌(MBC)疾病进展的理想替代标志物。近日,国际杂志 《CLIN CHEM》在线发表一项关于循环肿瘤细胞中的基因表达特征与转移性乳腺癌治疗的反应相关的研究。研究人员根据46个基因的表达情况调查了MBC患者CTC治疗反应的预测情况。 研究人员从45 MBC患者和20例健康供体(HD)中,在疾病进展时间(TP0)和2个临床分期时间点(TP1和TP2)收集2×5 mL血液采用AdnaTest EMT -2 / StemCellSelect TM(QIAGEN)进行检测。患者在TP1分组为(a)应答者(R)和无应答者(NR),在(b)TP2分组为总反应者(OR)和整体无应答者(ONR)。PCR检测46基因用于预扩增和高通量基因表达谱分析。使用GenEx(MultiD)和SAS分析数据。 CTC阳性由四基因特征(EPCAM,KRT19,MUC1,ERBB2阳性)决定。研究14个基因在CTC +和CTC-患者(KRT19,FLT1,EGFR,EPCAM,GZMM,PGR,CD24,KIT,PLAU,ALDH1A1,CTSD,MKI67,TWIST1和ER
