ACS Nano:美中澳科学家合作:利用“干细胞-生物材料”治疗心脏病

在世界范围内,心血管疾病是引起疾病死亡的主要原因,但是治疗手段仍然有限。心肌梗死可导致心肌的损伤,具有发病急,预后差的特点。近期,来自美中澳三地的科学家联合研究发现,利用纳米级的温敏材料包裹心脏干细胞,能够在心肌梗死心脏中展现出良好的修复作用。该结果已在大动物(猪)心梗模型中得到了验证,研究成果以Editors’ Choice的形式发表在《ACS Nano》杂志上。

研究学者目前已经尝试采用不同的方式修复损伤的心肌,而干细胞治疗是具有治疗潜力的方式之一。但是,单纯干细胞的治疗效果受限于细胞移植后滞留率低和宿主的免疫排斥反应等问题。在生物工程领域,由水和聚合物合成的可注射水凝胶在梗死心脏中的修复作用正在得到深入的研究。但是,天然的聚合物费用昂贵,各个批次之间差异性较大。

来自于美国北卡罗来纳州立大学和北卡罗来纳大学-教堂山分校的程柯教授、 中国郑州大学第一附属医院的张金盈教授和澳大利亚阿德莱德大学的Hu Zhang教授带领三方课题组联合研发孔径极小的纳米级P(NIPAM-AA)温敏材料,利用P(NIPAM-AA)温敏胶的物理特性和化学特性,与心脏干细胞结合,观察其在心肌梗死心脏中的修复作用。

研究发现P(NIPAM-AA)温敏胶在温度达到将近体温时可以由液态转为胶态。基于P(NIPAM-AA)温敏胶的特殊结构,这种廉价的合成水凝胶可以封装心脏干细胞,为干细胞提供良好的生长环境,并且不影响干细胞生长因子的释放。将封装的心脏干细胞注射入小鼠或者猪的体内,干细胞在体内的滞留率远大于单纯细胞注射组。P(NIPAM-AA)温敏胶的物理特性可以隔离心脏干细胞,一定程度上避免系统性炎症反应和局部免疫排斥反应的发生。在小鼠和猪的心肌梗死动物模型中的研究证实P(NIPAM-AA)温敏胶结合心脏干细胞具有良好的心脏修复作用。因此,P(NIPAM-AA)温敏胶封装干细胞治疗的方式克服了既往干细胞治疗的难题,全方位促进心脏干细胞长期有效的发挥心脏修复作用。

澳大利亚阿德莱德大学Hu Zhang教授,现任澳大利亚阿德雷德大学化工系高级讲师。主要研究方向为再生医学和组织工程,合成生物技术和生物工艺和过程模拟和计算流体力学。
