CLIN CANCER RES近期发表了一篇文章，研究索拉非尼联合抗炎性止疼药治疗难治性RAS突变肿瘤的效果。

作者在体外利用RAS突变细胞系研究乙酰水杨酸（阿司匹林）联合多酶抑制剂索拉非尼（Nexavar）的细胞毒性。使用选择性抑制剂或shRNA敲除基因来研究细胞毒性增强的机制。在RAS突变异种移植鼠模型中验证体外实验的结果。研究结果表明，阿司匹林可以显着增加索拉非尼体外的细胞毒性，而布洛芬和塞来昔布均无此效果。机制上看，联合暴露导致BRAF/CRAF二聚体增加同时刺激AMPK和ERK通路高度活化。索拉非尼和其他AMPK激活剂如二甲双胍或A769662，由于单独激活AMPK通路，降低细胞活性的程度不足。索拉非尼和阿司匹林的细胞毒性可以通过shRNA或药物抑制剂RAF（LY3009120）抑制AMPK或ERK通路进行阻断。两种药物的联合效应对RAS/RAF突变细胞具有特异性，对RAS/RAF野生型细胞无显着作用。在鼠模型中索拉非尼和阿司匹林联合应用与单药相比可以显着降低肿瘤负荷。

文章最后认为，索拉非尼联合阿司匹林同时影响两个独立通路，对RAS/RAF突变细胞具有毒性作用，在治疗RAS突变肿瘤方面具有前景。
