SCIENCE:肿瘤错配修复缺陷影响抗PD-1免疫疗法效果

这种高突变负荷使肿瘤具有免疫原性并对程序性细胞死亡-1(PD-1)免疫检查点抑制剂敏感。然而,尽管它们具有肿瘤免疫原性,但MMR缺陷型肿瘤患者的反应变化很大,大约一半的患者难以治疗。

具有错配修复缺陷(MMR-d)的肿瘤的特征在于微卫星的序列改变,并且其可累积数千个突变。

这种高突变负荷使肿瘤具有免疫原性并对程序性细胞死亡-1PD-1免疫检查点抑制剂敏感。然而,尽管它们具有肿瘤免疫原性,但MMR缺陷型肿瘤患者的反应变化很大,大约一半的患者难以治疗。

最近,研究人员提出的实验和临床证据表明,微卫星不稳定性(MSI)和所产生的突变负荷的程度部分地构成了MMR-d人和小鼠肿瘤中对PD-1阻断免疫疗法的可变反应的基础。响应程度特别与插入 - 缺失(插入缺失)突变负荷的累积有关。

该研究为MSI强度和突变负荷的全基因组表征提供了理论基础,以更好地描述对MMR缺陷型人类癌症的抗PD-1免疫疗法的反应。

痛风致腕关节破坏误诊为肿瘤1例
