Circulation:gp130调控心肌细胞增殖和心脏再生

心血管疾病始终是死亡的首要原因,心血管疾病高死亡率的一个关键原因是心肌细胞损伤后不能再生。心脏再生是降低心血管死亡率的有效方法,但真正实现心脏再生治疗,我们仍任到重远,需要不断探索。

一直以来,血管疾病始终是死亡的首要原因。心血管疾病高死亡率的一个关键原因是心肌细胞损伤后不能再生。在生理学上,心肌细胞是唯二之一的永久细胞(另一个是神经元细胞),即在个体出生后不会再分裂增殖。

心脏再生,是一种有前途的可修复损伤心脏的策略,即人为补充有功能的心肌细胞。然而,我们距将心脏再生应用于临床还有很长的一段路要走。首当其冲,我们需要先了解心脏再生的细胞分子机制。近日,《Circulation》上发表的“gp130 Controls Cardiomyocyte Proliferation and Heart Regeneration.”一文,揭示了gp130在心脏再生中的作用及分子机制。

该研究采用了7种不同基因编辑的小鼠:抑素M(OSM)全敲、单核细胞/巨噬细胞特异性OSM敲除、心肌细胞特异性品系(包括OSM受体敲除、gp130敲除、gp130激活和Yap废除+gp130激活);和两种心肌损伤模型:根尖切除术和心肌梗塞手术。

通过RNA测序发现,OSM是新生鼠心脏再生过程中心肌细胞增殖的关键上游调控因子。当OSM在巨噬细胞中被条件性敲除时,新生鼠心肌损伤后的心肌细胞增殖和心脏再生受到抑制。心肌细胞特异性OSM受体异二聚体、OSM受体和gp130缺陷可分别减少心肌细胞增殖和新生鼠心脏再生。

条件性激活心肌细胞的gp130可促进少年鼠和成年鼠的心肌细胞增殖和心脏再生。通过RNA测序和功能性筛查,研究人员发现Src通过Y457位点磷酸化激活Yap介导gp130触发的心肌细胞增殖,且独立于Hippo通路。在Myh6-gp130ACT小鼠中特异性敲除心肌细胞的Yap可阻断少年鼠的gp130激活诱导性心脏再生的效应。编码组成性激活型gp130的腺病毒相关病毒基因疗法可促进成年鼠心肌梗死后的心肌细胞增殖和心脏再生。

总结:巨噬细胞招募是通过分泌OSM在心脏再生过程中发挥至关重要的作用;OSM可促进心肌细胞增殖。在心肌再生过程中,gp130作为OSM的受体,其激活后可通过Src激活Yap有效促进心肌细胞增殖。总之,gp130是提高心肌损伤后心肌再生的有潜力的治疗靶点。
