Clin Chem:基于连接和模板转换的小RNA文库制备方案对生化酶的比较分析

RNA是调控基因表达和分化的关键分子。然而,许多不同种类的小RNA (sRNAs)具有不同的生物发生途径,因此可能具有不同的生化特性。为了通过深度测序分析sRNAs,互补DNA合成需要对RNA分子本身进行操作。因此,由于生化标准的存在,建文库过程中的酶活性会对文库的内容产生偏差。

我们比较了四种不同的RNA制备文库操作:(a)基于连接的方法仅允许5'-单磷酸化RNA进入文库,(b)基于连接的方法允许额外的5'-三磷酸和Cap结构 ,(c)基于连接的,基于模板转换的文库制备,和(d)基于模板转换的库制备,使3'-磷酸化的RNA进入文库。

我们的数据显示,依赖连接和依赖连接的文库在它们对单个sRNA类(如microRNAmiRNA),Piwi相互作用RNApiRNA)和转移RNA片段)的偏好方面存在很大差异。 此外,两种方法的miRNA组成不同,经过焦磷酸酶处理后可以识别出更多的microRNA亚型(isomiRs)piRNAs在模板转换库中得到了丰富,这一过程显然包含了更多不同种类的RNA

我们的数据表明,两种方法的miRNA组学几乎没有可比性。基于连接的文库丰富了典型miRNA,可能是适合miRNA组学研究的方法。模板转换库包含数量增加的片段和长RNAs的不同组成。连接无关的文库似乎显示了一个更真实的sRNA表达谱,对生化修饰的偏倚更低。
