摘要:

目前尚不清楚为什么激动剂刺激血小板需要剪切力才可有效地将促凝剂磷脂酰丝氨酸(PS)外化并释放PS-暴露的细胞微泡(MVs)。Aiming Pang等人发现整合素外-内信号是该要求的一种重要机制。

PS暴露和MV释放在β3-/-血小板或整合素拮抗剂的作用下受抑制。β3-/-血小板受损的MV释放和PS暴露可通过表达野生型β3得到恢复,但表达Gα13结合缺陷型β3突变体(E733EE-AAA)而不能,因该突变体阻碍外-内信号,而非配体结合。

抑制Gα13或Src也可削弱激动剂/剪切-依赖的PS暴露和MV释放,进一步提示了整合素外-内信号的作用。通过添加整合素配体增强拉力可诱导活化的血小板中的PS暴露,该效应可被抑制Gα13-整合素相互作用而消除,提示通过整合素作用的GGα13依赖性的机械信号传递可刺激PS暴露。

在体外,抑制Gα13会延迟凝血。而且,抑制或血小板特异性敲除Gα13可减少激光诱导的活体动脉血管内纤维蛋白形成。

综上所述,β3整合素作为剪切感受器,激活Gα13依赖的外-内信号通路,促进血小板的促凝血功能。药理靶向Gα13整合素相互作用可通过抑制凝血和血小板血栓形成预防活体内形成闭塞性血栓。
