PLOS ONE:男性肥胖对精子和下一代脐带血DNA甲基化的影响

人类和动物模型的流行病学研究表明,与不利的父母和/或宫内因素相关的表观遗传变化可能导致代谢紊乱的遗传缺失。可能不利的父系效应很可能通过精子传递给下一代。

全世界代谢紊乱,特别是肥胖症,的患病率急剧增加。遗传变异仅解释了由缺乏运动和过度营养引起的肥胖流行的一小部分。

人类和动物模型的流行病学研究表明,与不利的父母和/或宫内因素相关的表观遗传变化可能导致代谢紊乱的遗传缺失。可能不利的父系效应很可能通过精子传递给下一代。

为了研究这一假设,最近,研究人员系统地分析了男性体重指数(BMI)对精子表观基因组的影响,及其与下一代胎儿脐带血(FCBDNA甲基化的关系。

在接受体外受精或卵胞浆内单精子注射的294名人类供体的精子样本中,以及在所得后代的113FCB中,母体印迹(PEG1PEG4PEG5PEG10),父本印迹(H19-IG DMRIGF2-DMR0MEG3-IG DMR)区域,以及肥胖相关的非印迹HIF3A基因的甲基化水平通过亚硫酸氢盐焦磷酸测序来量化。

多变量回归分析显示,MEG3基因间差异甲基化区域(IG DMR)显示精子甲基化与供体BMI之间呈正相关。对于MEG3-IG DMRHIF3AIGF2-DMR0,观察到父本BMIFCB甲基化之间的性别特异性相关性。前两个基因在雄性后代的父本BMIFCB甲基化之间显示出相同的定向名义关联(如精子)。

随着父亲BMI增加,研究人员在女性后代的FCB中观察到IGF2-DMR0的低甲基化。

因此,该研究结果表明,男性肥胖名义上与人类精子DNA甲基化组的修饰有关,这可能会影响下一代的表观基因组。然而,重要的是要注意,没有相关的p值在多次测试调整中存活。未来的工作应该测试后代中相关甲基化畸变的影响,因为DNA甲基化被证明可以控制所研究基因的表达和/或印记建立。
