Blood:FXII和激肽原与gC1qR/C1QBP/P32的不对称组装受变构调控

在复合物中,XII因子纤连蛋白II型结构域与gC1qR的晶体结构表现为不对称的相互作用,并结合Zn2+离子;gC1qR将XII因子和激肽原簇聚合成更高阶的三元复合物。

接触系统,又称为血浆激肽释放酶-激肽系统,由因子XII (FXII)、激肽释放酶原(PK)和辅因子激肽原(HK)组成。球状C1q受体(gC1qR)已被证明可与FXII和HK相互作用。

Kaira等人发现FXII纤连蛋白II型结构域(FnII)以Zn2+依赖性的方式与gC1qR结合,决定了复合物的晶体结构。FXIIFnII与gC1qR的三聚体以非对称的方式结合,Arg36和Arg65残基与两个不同的带负电荷的口袋形成接触。
 
gC1qR的残基Asp185和His187都定位在FXII结合位点附近的一个Zn2+,与配体游离的gC1qR晶体结构比较发现,阴离子G1-环在FXIIFnII结合后变为有序排列。Zn2+结合位点区域的构像变化揭示了Zn2+调节FXII结合的变构基础。

结合SPR的诱变表明,gC1qR Zn2+位点有助于FXII结合,基于血浆的检测显示,gC1qR以FXII依赖的方式刺激凝血。HK 结构域 5 (HKD5)与gC1qR的结合分析显示每个三聚体只有一个高亲和力的结合位点。
 
诱变研究发现了一个位于gC1qR三聚体中心的关键的G3-环,揭示了空间闭塞是HKD5不对称结合的机制。凝胶过滤实验表明,gC1qR将FXII和HK结合成更高阶的500kDa的三元复合物。

总而言之,本研究结果支持了细胞外gC1qR可作为接触因子聚集的伴侣的结论,这可能是启动驱动缓激肽生成的级联反应和凝血内在途径的前奏。
