有3位诺贝尔奖得主对新冠mRNA疫苗进行了点评。他们分别是:2013年诺贝尔生理学或医学奖得主兰迪•谢克曼(Randy Schekman),1987年诺贝尔化学奖得主让-马里•莱恩(Jean-Marie Lehn),2004年诺贝尔化学奖得主阿夫拉姆•赫什科(Avram Hershko)。

三位诺奖得主都提到了mRNA疫苗技术的快速和高有效性。对于mRNA技术,他们表示,除了针对新冠病毒,它还有更为广泛的治疗潜力,可以说开创了一个用于更广泛疾病治疗的“mRNA时代”。

mRNA疫苗是一种通用的平台型技术,有望颠覆传统疫苗研发路径,同时将来应用前景也十分广泛,不仅仅是病毒或细菌等感染性疾病的预防,还有望用于癌症等诸多领域。与传统疫苗相比mRNA 疫苗主要有以下核心优势:(1可以表达任意种类的蛋白如天然蛋白抗体病毒蛋白等突破现有专利以及毒株菌株等方面的限制;(2可以同时表达多个蛋白有望开发出多价苗联苗等疫苗;(3可以模拟病毒自然感染人体的过程引起较强的体液免疫与细胞免疫但安全性较高且可以通过成熟的制剂技术如核苷酸修饰等调整核酸疫苗表达抗原的能力来调整体液免疫和细胞免疫之间的平衡;(4生产周期较短3 个月内即可完成 GMP 生产及 QC可以应对大规模突发性传染病;(5研发成功后可以快速开发出同一给药系统的更多系列产品为通用的平台型技术;(6标准化生产且场地要求低化学合成核酸普通 GMP场地即可安全性高且不易因批次间的变化而造成不必要的批次损失;(7共线生产所有产品既不涉及病原体也不涉及细胞培养或发酵体系因此一个单一的设施可以生产所有的 mRNA 疫苗共享同一套质量体系生产设备以及操作人员综上所述mRNA 疫苗有望颠覆传统疫苗研发路径开启疫苗的新时代。

mRNA疫苗目前在黑色素瘤,囊性纤维化等领域都取得不错的进展。Moderna和BioNTech的目前正在研发的项目:其中包括治疗乳腺癌、前列腺癌、皮肤癌、胰腺癌、脑癌、肺癌和其他组织的药物试验,以及预防流感、寨卡病毒和狂犬病的疫苗。

COVID-19大流行释放了世界各地生物医学科学力量来设计疫苗,以阻止致命SARS-CoV-2病毒的传播。在中国、北美和欧洲,已设计出新方法来利用冠状病毒或其他形式的重组病毒大规模开发SARS-CoV-2疫苗,以便在2020年年底之前保护整个世界。

在生产传统病毒疫苗的同时,在美国和德国的公司中,一种改良基于编码SARS-CoV-2刺突蛋白的RNA疫苗,在开发了数年后现在已首次商业化。与注射蛋白质抗原以引发免疫反应的传统疫苗不同,新方法中使用的刺突蛋白信使RNA(mRNA)是化学合成的,然后配制成脂质纳米颗粒,以便在注射部位传递到肌肉细胞中。传统疫苗会与我们体内的免疫细胞发生反应,其中一些在早期发育过程中也被偶然地预编为与异源抗原(例如,表面抗原)表面上的化学基团,SARS-CoV-2刺突蛋白表位发生反应,随后刺激与表位反应的免疫细胞生长和分裂,并发展成抗体产生细胞以及在将来暴露于外源抗原(即感染)的情况下充当贮存器的记忆细胞。在新技术中,脂质纳米颗粒进入细胞后剥离,然后通过暴露的mRNA重新编码生产蛋白质的细胞机制,以生产和输出SARS-CoV-2穗状蛋白。一旦重组刺突蛋白从这些细胞中分泌出来,免疫系统就会像传统疫苗方法一样接管。这种方法的优势在于,疫苗生产的整个过程都依赖于化学和工程技术,与用于生产更多传统病毒疫苗的生物过程相比,化学和工程技术可以得到更精确的控制。新的mRNA疫苗提供了几乎完全的保护,这是一个优势。毫无疑问,我们会将此技术应用于其他医学治疗。
