7月3日,国际学术期刊《美国国家科学院院刊》(PNAS)在线发表了中国科学院上海营养与健康研究院计算生物学研究所韩敬东研究组发表的论文Repression of Human and Mouse Brain Inflammaging Transcriptome by Broad Gene Body Histone Hyperacetylation。该研究揭示了表观遗传修饰对大脑炎性衰老基因调控的新模式,表明了炎性衰老进程的可逆性,为干预衰老相关的大脑功能下降以及神经退行性疾病提供了新视角。

大脑的炎性衰老是衰老相关的神经退行性疾病的一个主要标志。研究人员通过对人和小鼠大脑中H3K27ac和基因表达数据的整合分析,发现随衰老上调和下调的基因上具有完全不同的H3K27ac修饰模式。尽管H3K27ac在两类基因的启动子上均有分布,然而富集免疫相关功能并随衰老上调的基因的基因体上却有着更为广泛的H3K27ac修饰,并且这些修饰会随着衰老逐渐丢失。与启动子区H3K27ac的转录激活功能不同,基因体超乙酰化抑制了炎性衰老相关基因的过度激活,并且基因体上的H3K27ac水平,可以用来预测基因在衰老过程中的变化方向。上述研究证实了H3K27ac在随衰老上调或下调的基因上行使截然相反的调控功能,并且提出了基因体超乙酰化抑制转录的新模式。
