肿瘤相关白细胞增多症(TRL)与各种类型的癌症的生存能力差相关,但TRL相关的人类肿瘤的微环境尚未完全阐明。在此,我们旨在描述TRL相关癌症患者的免疫微环境。

研究比较了从宫颈癌患者中获得的肿瘤组织的转录特征(TRLpos)和无TRL(TRLneg)的肿瘤组织的转录特征。作为TRL诊断的代用品,使用TRLpos和TRLneg肿瘤之间表达不同的基因,得出了白细胞增生特征(LS)评分。比较了TCGA数据库中高(LShigh)或低LS评分的患者的免疫学谱。

 

结果显示,TRLpos肿瘤与TRLneg肿瘤在转录学上存在差异,表现出上调放射抗性和下调适应性免疫反应相关基因。在TCGA宫颈癌队列(n = 303)中,高LS的患者与低LS的患者相比生存率较低(P = 0.023)。LShigh肿瘤在放射抗性、伤口愈合和髓样抑制细胞(MDSC)标志物中含量较高,M2巨噬细胞浸润度较高,M1巨噬细胞和淋巴细胞浸润度较低。LShigh肿瘤也表达了更高的CXCR2趋化因子、CSF2和CSF3。在泛癌队列(n = 9984)中,LShigh肿瘤也表现出较差的存活率、抑制性免疫微环境的特征和CXCR2趋化因子的较高表达。

 

总之,我们的数据为TRL患者的抑制性免疫微环境提供了证据,并提出了有前景的靶点,如CXCR2轴,对其进行治疗干预。
