近日,发表在Nano Today杂志上一项研究,研究人员开发了一种纳米酶,可以阻止错误折叠的α-突触核蛋白的扩散,并可能为开发PD新疗法奠定基础。这种纳米酶是由铂和铜组成的PtCu合金纳米酶,具有良好生物兼容性,且拥有强大的抗氧化性。纳米酶是一类可模拟生物酶催化活性的纳米材料,具有很多天然酶不可比拟的优势,至今,科学家们已发现数百种有类酶催化特征的纳米酶。

研究人员通过α-突触核蛋白预制纤维体(PFF)模型的研究方法,该模型复制了路易体(LB)诱导的帕金森疾病的病理学和神经病变。在神经元细胞培养物中添加PFF后发现ROS水平升高,α-突触核蛋白积聚,呈现出病理性和神经毒性。然后,加入PtCu合金纳米酶后,不仅能显著降低ROS水平,降低α-突触核蛋白病理性和神经毒性,更为重要的是能阻断病理性α-突触核蛋白在细胞间的传播,以及从纹状体向黑质的扩散。

越来越多的研究表明,变形和错误折叠的α-突触核蛋白是导致帕金森病的罪魁祸首,它从肠道传播到大脑,在大脑中扩散并粘在一起,形成致命的团块。研究人员希望能进一步开展针对肠道与帕金森病的研究,为帕金森病治疗提供新的治疗策略。总之,PtCu合金纳米酶在预防散发性PD中的α-突触核蛋白的扩散表现出显著的疗效,为帕金森病治疗提供了新策略。
