今天就来讲一下,衰老对免疫细胞功能的影响。

衰老是每个人都不得不面对的现实,随着年龄地增加,衰老带来的不仅仅是容貌的改变,人的健康状况也会随之下降,各种慢性疾病开始不断地入侵。   衰老是癌症最大的诱因。衰老引起的最明显的现象是免疫系统的失调,包括免疫衰退和慢性炎症。免疫系统的衰退削弱了针对肿瘤细胞和病原体的抵御能力,同时慢性炎症状态增加了自身免疫性疾病的风险。   随着年龄的增加,癌症发病率逐渐升高,现代科学发现其中最关键的一点为:衰老导致了多种免疫细胞的功能下降。   

那么,我们今天就来讲一下,衰老对免疫细胞的影响。

衰老对T细胞的影响

T细胞是免疫细胞的主要组成部分,它具有多种免疫功能,如产生细胞因子、参与特异性免疫应答、清除病原体、衰老细胞、突变细胞以及癌细胞,是人体中为抵御疾病感染、肿瘤而生的警卫队。   因此,   T细胞的数量功能及多样性对抵御癌症至关重要。   研究发现,儿童时期,随着人体接触到的病原体种类越来越多,体内T细胞的多样性也逐渐升高,并在20岁左右到达最高点。      25岁后,随着年龄的增加,T细胞的多样性随之下降。

与细胞多样性发展趋势相反的则是疾病发生率。T细胞的多样性越高,则意味着能够杀灭各种细胞病原体的种类越多,   随着年龄的增加,细胞多样性降低,无法消灭相应的病原体,疾病发病率也就随之增加   。

衰老对B细胞的影响

B细胞又称B淋巴细胞,来源于骨髓多能干细胞。成熟的B细胞主要定居于淋巴结皮质浅层的淋巴小结和脾脏的红髓和白髓的淋巴小结内。B细胞参与人体的体液免疫,在抗原刺激下可分化为浆细胞,浆细胞可合成和分泌抗体(免疫球蛋白),杀灭病原体及细胞。

但是研究发现,衰老也会影响B细胞的质量和数量。与年轻人相比,老年人的B细胞百分比数量下降,抗体类别转化重组缺陷。不仅如此,记忆B细胞数量增多,导致了老年患者慢性炎症的发生。

随着年龄的增长, 老年人的免疫效果明显下降。面对相同的病原体,发挥作用的B细胞数仅及年轻时的1/10~1/50 。例如,接种流感疫苗后60~74岁组的血清阳性保护率为41%~58%,75岁以上的阳性保护率下降到29%~46%。

衰老对NK细胞的影响

自然杀伤细胞(natural killer cell,即NK细胞)是机体重要的免疫细胞,不仅与抗肿瘤、抗病毒感染和免疫调节有关,而且在某些情况下参与超敏反应和自身免疫性疾病的发生,能够识别靶细胞、杀伤介质。   同时,在面对癌细胞时,NK细胞不需要其他细胞的激活与刺激,便能直接识别并杀死癌细胞,因此也被称为人体抗癌第一道防线。   然而研究发现,   NK细胞的数量及功能在40~50岁后显著下降。NK细胞的功能逐渐减弱,癌症发病率却随之增加。   在对日本中年及老年人随访11年后显示,NK细胞的活性和功能与肿瘤组织的大小、转移以及不良预后息息相关。

不仅如此,随着年龄的增加,老年人群体内的免疫调节细胞数量逐渐增加。与T细胞及NK细胞不同的是,免疫调节细胞通过抑制免疫细胞发挥,维持体内免疫平衡。然而在老年人体内,过多的免疫调节细胞却抑制了NK 细胞发挥作用。   随着医学的不断发展,我们逐渐认识到衰老、疾病的发展、甚至癌症的产生都与免疫细胞有着千丝万缕,不可分割的关系。对于癌症,我们有了激活或增强免疫细胞,来消灭癌症的免疫疗法,拯救了无数的癌症患者。   

相信有朝一日,我们也能利用免疫细胞放慢衰老的脚步。
