这篇题为《胜负经历重塑丘脑到前额叶皮层环路以调节社会竞争优势》(History of winning remodels thalamo-PFC circuit to reinforce social dominance)的文章,由博士生周亭亭,朱鸿和范郑晓等在胡海岚教授的指导下共同完成。


翻盘的竞争

有人的地方就有江湖,其实这句话只说对一半。鱼类、鸟类、灵长类等动物中间也有“江湖”,科学家称之为社会等级。上世纪20年代,一位挪威的科学家最早在家鸡中观察到:等级高的鸡总是啄咬等级低的,而相反的状况则极少发生。


社会等级与我们的个体健康等息息相关,背后有什么神经机制? 2011年,胡海岚团队在《科学》发文,引入“钻管测试”来研究小鼠的等级地位:在一段只能让一只小鼠通过的玻璃管道中,两只小鼠狭路相逢,一场不进则退的较量在所难免,而优势者会在30秒内将对方推出管道。一群小鼠经过两两竞争,等级高低便一目了然。


当年这项研究最激动人心之处,还在于第一次向世界指出了调节社会竞争的神经基础,位于大脑内侧前额叶的脑区。当科学家定向增强内侧前额叶脑区的突触(大脑细胞之间连接、通讯的基本结构单元)强度,处于劣势的小鼠就像服用了“大力神丸”,勇气倍增,有如神助地将优势小鼠逼出玻璃管道,成功逆袭。

重复成功塑造大脑


基于上述发现,利用光遗传学的方法(一种利用激光来瞬时、特异激活神经元的手段),胡海岚团队在这项研究中进一步实时“操纵”了钻管竞争的输赢。并且发现,小鼠等级地位相差越悬殊,逆袭所需的“神经激活剂量”就越高。但她们并没有停止研究。团队成员发现了更为有趣的现象:

当劣势小鼠成功逆袭6次或更多次时,即使离开科学家的“帮助”,“裸奔”的小鼠依然能实现逆袭。而成功经历不足6次,裸奔的劣势小鼠会恢复到劣势地位。“这种先前胜负经历影响后续比赛输赢的现象,恰恰体现了心理学中的‘胜利者效应’。”周亭亭说。

这一效应是如何产生的呢?团队发现了一个从中缝背侧丘脑投射到前额叶皮层神经通路,当增加这一环路突触连接的强度,就能介导“胜利者效应”。“成功经历会重塑这一通路的突触连接强度,从而影响后续竞争中的表现。”胡海岚说。

也就是说,在胜利了6次之后,劣势小鼠的大脑发生了质变,他们的神经突触连接强度显着增加了,有效地帮他们进阶并维持在更高的等级地位。

研究团队认为,这是一条在进化上保守的神经环路,从低等哺乳动物到高等哺乳动物乃至人类中普遍存在,因此会有更多的启示意义。

“强者恒强”

为了验证这一“胜利者效应”,研究团队还设计了一项热源争夺战的实验。在一个冰冷的方形盒子中,四只小鼠对位于盒子角落的温暖地带展开竞争。只要是之前在钻管测试中获得重复胜利经历的小鼠,在热源争夺中也会更容易获胜。周亭亭认为,这一结果首次说明“胜利者效应”可以从一种行为学范式迁移到其他的行为中。

《科学》杂志的评审专家认为,“这是一篇杰作,它运用了多种技术手段实现了对神经回路的操控,向我们展示了令人惊异的而且清晰的行为现象及行为范式。”学术界认为,这项研究为研究社会等级的形成和稳定提供了新的思路和研究方法,也为对输赢决定、社会等级的认知等的神经环路进行更为细致的研究提供了新的靶点脑区。


“胜利者效应”是一种重要而有趣的现象,都柏林大学圣三一学院心理学教授伊恩?罗伯逊曾着书《胜利者效应》,讲述了从动物到人类世界发生的各种“胜利者效应”的故事,其中就有拳王泰森的一段:泰森锒铛入狱数年后复出,经纪人巧妙设计了两场比赛,对手实力远低于泰森。不出所料,泰森赢得了这两场比赛。紧接着,泰森才战胜了强大的对手布鲁诺,再次获得金腰带,重新走上事业巅峰。

朱鸿说,这项研究在动物实验中第一次验证“强者恒强”的现象并且阐释了其神经科学原理。虽然人类社会远比动物社会复杂,但是这项研究对于提高人们在各项比赛、考试、人际交往中的表现,也提供了有益的线索。

胡海岚教授长期致力于研究情绪与社会行为的分子与神经环路机制,曾先后在冷泉港实验室和中科院上海生命科学研究院从事研究工作,2015年5月加入浙大。中科院院士,医学部主任段树民说,学校尽可能充分地、灵活地调动各方资源,为科学家营造良好的研究环境。段树民认为,脑的高级认知、情感等行为的神经环路和神经生物学机制是脑科学中最具有挑战性的问题,胡海岚在该领域有着系统的创新性工作,受到国内外同行的广泛认可。“看到胡海岚团队最近又取得既有理论意义又有潜在应用价值的原创成果,由衷为她的团队感到高兴。”

据介绍,该项研究受到浙江大学985基金、科技部973项目,中国科学院先导计划B,神经科学国家重点实验室、国家杰出青年基金等资助。
