摘要:

黏连蛋白复合体破坏会改变基因的表达,在髓样瘤变中常见黏连蛋白突变,提示黏连蛋白在造血过程中起着重要作用。Daniel Sasca等人对黏连蛋白的动力学以及对造血干细胞的稳态和分化的调控作用进行研究。

之用在红细胞分化过程中,活性调控元件上结合的黏连蛋白才回增加。抑制性Ets转录因子Etv6的预结合可预测这些元件的黏连蛋白结合,以及Etv6与黏连蛋白在染色质上的相互作用。敲除黏连蛋白可严重破坏红系分化,特别是Etv6预结合的位点,但会增强自我更新的程序。

综合在急性髓系白血病和骨髓增生异常综合征患者样本中的研究结果,上述数据表明,黏连蛋白介导的Etv6抑制的缓解是维持红系细胞分化过程中关键的红系基因动态表达所不可缺少的。
