JEM:中科院阐明CCP6蛋白调控血液巨核细胞谱系发育机制

2014年10月20日,Journal of Experimental Medicine在线发表了中科院生物物理所范祖森和田勇等人题为“Cytosolic carboxypeptidase CCP6 is required for megakaryopoiesis through modulating Mad2 polyglutamylation ”的研究结果,这一工作首次探明了胞内羧肽酶CCP6及其调控的Mad2蛋白的多聚谷氨酸化修饰对造血谱系发育中骨髓巨核细胞成熟具有的调控作用。

众所周知,血液是维持机体生理功能重要的组成部分。但是,血液细胞谱系建立的机制到现在仍未完全揭示。骨髓巨核细胞是血小板发育的前体细胞,关于骨髓巨核细胞生成、发育及成熟分化的分子机制目前阐释的并不清楚。范祖森与田勇等人利用基因敲除小鼠模型系统探讨了羧肽酶CCP6蛋白对骨髓巨核细胞定向发育调控的分子机制。研究发现,胞内羧肽酶CCP6的基因敲除小鼠体内血小板增多、功能异常、骨髓巨核细胞增生,脾内也出现明显的巨核细胞增生灶。CCP缺陷小鼠巨核细胞在超微结构观察中呈现明显的不成熟特征,DNA倍性低,成熟障碍。通过Pulldown及质谱分析鉴定了有丝分裂纺锤丝检查点蛋白Mad2可以与CCP6相互作用。CCP6的基因敲除使Mad2蛋白的多聚谷氨酸化修饰程度增加。研究鉴定了巨核细胞中,调控添加多聚谷氨酸化酶TTLL4及TTLL6高表达,在体外重组的谷氨酸化修饰体系中,TTLL4/6可以为Mad2加谷氨酸化修饰,CCP6可以去除Mad2蛋白的谷氨酸化修饰。进一步鉴定了Mad2蛋白发生谷氨酸化修饰的位点,通过移植实验研究观察到TTLL6及Mad2的谷氨酸化修饰调控了巨核细胞体内发育成熟过程。研究人员同时还发现,谷氨酸化修饰的Mad2结合Aurora B激酶的能力增强,CCP6的基因敲除促使Aurora B的磷酸化水平增强、活性提高。研究结果表明,机体内CCP6和TTLL6通过调节Mad2的谷氨酸化修饰,调控了巨核细胞的成熟以及血小板的产生。这一研究成果为揭示血小板的产生机制提供了重要的实验依据,为血小板的再生医学研究提供了重要的理论基础。
