GUT:新型小鼠模型为人特异性肿瘤微环境免疫疗法提供了试验条件

由于目前人类肝细胞癌(HCC)的治疗策略效果有限,免疫疗法成为解决该疾病的最有希望的一种治疗模式。本项研究的目标是构建人源化小鼠(humice)模型,以了解人类癌症和免疫系统之间的相互作用,为人特异性肿瘤治疗提供实验基础。

目的
由于目前人类肝细胞癌(HCC)的治疗策略效果有限,免疫疗法成为解决该疾病的最有希望的一种治疗模式。本项研究的目标是构建人源化小鼠(humice)模型,以了解人类癌症和免疫系统之间的相互作用,为人特异性肿瘤治疗提供实验基础。

方法:
在模型小鼠中用I型人白细胞抗原匹配的人免疫系统建立患者来源的异种移植肿瘤。通过长期观察研究了小鼠体内肿瘤的纵向变化和免疫应答以及免疫检查点抑制剂的功效。

结果
本项研究中的人源化小鼠模型中的类人体免疫系统受到肿瘤的影响,并表现出疲态现象,例如白细胞数量的显着下降,疲惫标志物的上调和人类促炎细胞因子的产生减少。值得注意的是,与其他细胞类型相比,细胞毒性免疫细胞减少得更快。与外周T细胞相比,肿瘤浸润的T细胞具有更高的耗竭标志物表达和更低的细胞因子产生。此外,研究人员发现肿瘤相关的巨噬细胞和骨髓衍生的抑制细胞在肿瘤微环境中高度富集。有趣的是,肿瘤还通过上调免疫检查点配体来改变响应免疫应答的基因表达谱。

结论
本项研究的成果为免疫肿瘤学研究提供了一个模型,并为抗HCC药物检测,特别是免疫治疗提供了一个有用的人源化平台。
