Sci Rep:外泌体在单纯性脂肪肝进展到NASH中发挥重要作用

非酒精性脂肪性肝病(NAFLD)是慢性肝病发生的主要病因,但是从单纯性脂肪变性到非酒精性脂肪性肝炎(NASH)的确切机制目前尚不清楚。本研究中研究人员探究了外泌体在NAFLD进展中的作用。研究人员使用棕榈酸(PA)分离人肝癌细胞株分离出外泌体,并使用miRNA芯片检测miRNA。然后使用从肝细胞分离的外泌体处理肝星状细胞(HSC)系(LX-2)。与对照组相比,PA处理的肝细胞CD36得表达水平显著

研究人员使用棕榈酸(PA)分离人肝癌细胞株分离出外泌体,并使用miRNA芯片检测miRNA。然后使用从肝细胞分离的外泌体处理肝星状细胞(HSC)系(LX-2)。与对照组相比,PA处理的肝细胞CD36得表达水平显著升高,且有外泌体的产生。微阵列分析显示,来源于赋形剂与来源于PA处理的干细胞的外泌体内miRNA的表达存在明显的差别。与使用赋形剂处理肝细胞的外泌体与LX-2细胞共培养相比,使用PA处理肝细胞的外泌体与LX-2细胞共培养时,与肝细胞纤维化发生相关的基因表达明显增多。

总之,该研究表明,PA处理肝细胞可显著增强Exosomes的产生,并会改变外泌内miRNA的表达谱。此外,来自PA处理的肝细胞的外泌体可引起HSCs内纤维化基因表达水平的增加。因此,外泌体可能在单纯性脂肪肝进展到NASH的过程中起着非常重要的作用。
