PNAS:先天愚有治疗希望了!科学家找到改善唐氏综合征的药物

唐氏综合征是人类最常见的染色体异常,大约每1000个人中就有一个。它的特点是大脑功能下降。当一个人有21号染色体的额外拷贝时,就会出现神经发育问题。在这项新的研究中,研究人员从寻找一种能使干细胞成长为神经细胞的化合物而开展他们的研究工作。在研究了717种可能性候选药物之后,研究小组找到了一种他们称之为ALGERNON(altered generationof neurons)的化合物。作为他们研究的一部分,他们研究了这种化合物如何影响已知患有唐氏综合征的小鼠胚胎。

研究小组通过将化合物添加到怀孕小鼠的饲料中,来研究这一化合物对小鼠胚胎大脑的影响。他们发现,化合物的使用会使大脑皮层正常发育(但在唐氏小鼠身上,结构比正常的要薄)。更好的是,他们发现,在出生到成年后小鼠的行为是正常的,在解决迷宫问题上和正常的小鼠一样。

研究小组接下来在培养皿中将这个化合物加入到患有唐氏综合征患者的人类干细胞中,发现细胞生长正常,这一发现当然也提供了这种化合物防止人类大脑畸形的可能性。但是,研究小组指出,用人类胚胎进行测试还有很长的路要走。现在仍不知道药物可能对发育有其它什么影响,必须做更多的测试。研究小组指出,可能还有一些社会问题需要解决,因为在给孕妇服用药物时,医生实际上会从父母改变后代的身体特征。不过除了这些顾虑,ALGERNON有着巨大的研究潜力,不仅在治疗唐氏综合征上,而且有可能应用到许多神经发育障碍上。
