黑素瘤相关抗原家族A（MAGE-A）属于癌睾丸抗原家族，其在正常组织中几乎不表达，而在多种肿瘤组织中均有较高水平的表达，并且往往表达一种以上的MAGE-A亚型，作为一种肿瘤相关抗原，与肿瘤的发生、发展、耐药及较差的预后密切相关。

黑素瘤相关抗原家族A（MAGE-A）属于癌睾丸抗原家族，其在正常组织中几乎不表达，而在多种肿瘤组织中均有较高水平的表达，并且往往表达一种以上的MAGE-A亚型，作为一种肿瘤相关抗原，与肿瘤的发生、发展、耐药及较差的预后密切相关。目前的研究显示，MAGE-A定位于X染色体，共包括12个成员。p21基因是位于p53基因下游的细胞周期素依赖性激酶抑制因子，可以和p53共同构成细胞周期G1检查站，因DNA损伤后不经过修复则无法通过，减少了受损DNA的复制和积累，从而发挥抑癌作用。研究人员调查了MAGE-A12在肿瘤中的作用。研究结果显示，MAGE-A12在p21稳定性和肿瘤生长中起关键作用，表明MAGE-A12或为癌症治疗提供新靶标。调查Oncomine数据库和NextBio数据库显示，相对于相应的正常组织，恶性肿瘤组织显示MAGE-A12 mRNA上调。在PPC1原发性前列腺癌细胞和HCT116结直肠癌细胞（野生型和p53缺失型）中，细胞ATP水平、细胞计数和克隆形成试验显示，使用siRNA或shRNA敲除MAGE-A12基因减少癌细胞增殖。FACS分析Analyses V-PI染色和DNA
