长期以来,人们对有磁性的氧化铁纳米粒子(IONPs)已经广泛研究,但大多是把它们用于磁共振成像(MRI)中作为对照剂。直到最近,才开始探索把它们 用于药物递送。目前只有少数研究描述了怎样在磁性氧化铁纳米粒子的表面加载化疗药物,还不能有效证明它们真能把药物递送到细胞内部,只是一些推测。而本研 究设计了一种新方法在 IONPs 表面加载药物,并首次证明了这些粒子能把药物递送到细胞内部。

研究人员给 IONPs 装上一种精心设计的接枝聚合体外壳,使它在水中和血清中都显出极佳的胶质稳定性。聚合体外壳能通过亚胺键实现阿霉素(DOX,一种抗肿瘤药物)可逆性粘附,为 DOX 在酸性环境中提供了一种可控的释放机制。

利用一种称为“荧光寿命成像显微”(FLIM)的技术,研究人员首次证明了 IONPs 能很容易地被两种细胞系(MCF-7 乳腺癌细胞和H1299肺癌细胞)接受,同时可监控DOX在细胞内的释放。

研究人员指出,药物释放实验通常只是在实验室中模拟,而不是在细胞中。这一点很重要,用细胞科学家才能确定,在真正的生物环境中药物释放的动力运动情况。 这项研究证明了工作台化学(指传统的,不使用高端设备和计算机模型的化学实验)也能在细胞内进行,下一步就要进入活体应用了。
