炎症坏死(pyroptosis)对于宿主抵御感染及危险信号至关重要,但过度的炎症坏死会导致免疫性疾病和感染性休克。炎症坏死涉及细胞肿胀和溶解,导致大量释放细胞内容物,由此触发强烈的炎症。炎症坏死这一术语最初被用来描述caspase-1介导的巨噬细胞死亡。Caspase-1属于炎症相关caspase,炎症相关caspase还包括有小鼠caspase-11和它的人类对应物caspase-4及-5。不同于caspase-11,caspase-4和caspase-5还可在非单核细胞中激活炎症坏死。Caspase-4,-5和-11可感知并通过直接结合脂多糖(LPS)而被激活;这些caspase过度活化可导致感染性休克。

在2015年的一篇Nature文章中,邵峰课题组证实炎症caspase通过切割gasdermin D(GSDMD)蛋白控制了炎症坏死3。所有的炎症相关caspase均在它的N-末端gasdermin-N结构域与C-末端gasdermin-C结构域的分子内抑制。却是GSDMD不会影响caspase-1加工白介素(IL)-1β,但会阻止成熟IL-1β分泌,表明炎症坏死是非经典细胞因子分泌的必要条件。

除了GSDMD,gasdermin家族还包括GSDMA,GSDMB,GSDMC,DFNA5和DFNB59。小鼠没有GSDMB,但具有三种GSDMA(GSDMA1-3)和4种GSDMC(GSDMC1-4)蛋白。其他gasdermin队炎症相关caspases不敏感。GSDMA3和DFNA5显性突变(或DFNB59常染色体隐性突变)可在小鼠中引起脱发和角化过度,在人类中导致非综合征型耳聋。疾病相关GSDMA3突变体和它的gasdermin-N结构域由于丧失自抑制可激活炎症坏死。尽管gasdermin在炎症坏死和炎症中起重要作用,当前对于GSDMD和gasdermin家族的作用机制仍不清楚。

在这篇Nature文章中研究人员证实gasdermin蛋白GSDMD,GSDMA3和GSDMA的gasdermin-N结构域可以结合膜脂、磷酸肌醇和心磷脂,在哺乳动物细胞和人工转化细菌中显示出破坏膜的细胞毒性。在炎症坏死过程中gasdermin-N移动到质膜处。纯化的gasdermin-N可有效溶解包含磷酸肌醇/心磷脂的脂质体,在人工或自然磷脂混合物构成的膜上打孔。大多数gasdermin孔内部直径为10-14nm,这些分子孔道具有16重对称性。GSDMA3的晶体结构显示出保守存在于gasdermin家族中,gasdermin-N端结构域与C端结构域之间的精细的自抑制相互作用。结构指导的诱变研究证实了gasdermin-N结构域具有的脂质体泄露和打孔活性是炎症坏死的必要条件。

这些研究结果揭示出了炎症坏死的机制,提供了gasdermin家族在坏死、免疫和疾病中所起作用的一些重要新认识。
