目的：结直肠癌患者临床管理的大大改善是由于对新靶标如EGFR和VEGF的识别。因为肿瘤的快速增殖与患者的不良预后有关，这里我们确定快速增殖的结直肠癌细胞的转录信号特征，试图确定新型候选的治疗靶标。实验设计：对52个结直肠癌细胞系的倍增时间进行确定，采用微阵列分析对这些线性子集的全基因组表达进行评估。然后我们调研快速增长的肿瘤细胞潜在基因高表达的作为新的治疗靶标。结果：快速增长率与微标记物的不稳定性

结果：快速增长率与微标记物的不稳定性和低分化组织学相关。1290个基因的表达与结直肠癌细胞的增长率显著相关。这些基因包括参与细胞周期，RNA加工/拼接和蛋白质转运的。甘油醛3-磷酸脱氢酶(GAPDH)和原卟啉原氧化酶(PPOX)显示在快速增长的高表达细胞株和主要肿瘤中有高表达。药理或基于siRNA 抑制的GAPDH或PPOX能降低体外结肠癌细胞的生长。此外，使用小鼠异种移植模型，我们发现以特异的PPOX抑制剂三氟羧草醚处理能显著的降低3/7(42.8%)结肠癌细胞线性的增长。

结论：我们已经发现了在不同增长率结直肠癌细胞之间转录水平的差异，并确定用于结直肠癌治疗的新型候选的化疗靶标。
