质粒在cmv、cox2或者是opn启动子的控制下可编码分泌性报告因子荧光素酶(G.LUC)。研究人员通过聚乙烯亚胺将这些报告系统转化到培养基上的膀胱癌肿瘤细胞中和小鼠的膀胱中。研究人烟记录了报告子的表达情况,并确定了报告因子检测的最优时间,还评估了报告因子与肿瘤大小的联系。体外试验表明cox2和opn启动子可以以针对性的方式启动G.LUC在膀胱癌细胞中显著的表达。体内试验表明cox2启动子引起了G.LUC的表达,且在尿液中达到了可检测的水平,并且在转化后的48-72个小时能达到局部信号最大值。另外,研究人员还发现,G.LUC在尿液中水平具有24小时的周期性,周期性部分的归因于由肿瘤细胞分泌的一种物质,这种物质可以掩盖荧光素酶信号。最后,研究人员指出,该检测系统具有肿瘤特异性,并且具有标准化的昼夜表达模式,对泌尿系膀胱癌和其他癌症展现出未来检测巨大应用前景。
