事实上,去年第二军医大学干细胞与医学创新研究中心主任胡以平在Cell Stem Cell杂志上发表文章称,找到纤维细胞可转分化为肝干细胞的方法,该方法可为肝脏疾病治疗奠定了一个新的基础。

肝脏细胞在很长时间内保留再生能力,例如一个肝脏捐赠者,最多可捐赠75%的肝脏,而这之后,剩下的肝脏可再生至原来的摸样。有学者猜测,肝脏中或许有潜在的干细胞帮助修复温和的肝损伤。

该研究支持了这一理论。Hippo信号通路是一条细胞抑制生长性信号通路,能调节细胞增殖和凋亡,帮助器官在生长结束后停止继续生长。目前研究人员正在研究Hippo信号通路是否会影响细胞的命运。

“自90年代初,人们就一直在试图利用肝细胞移植来治疗代谢疾病,然而由于肝细胞是来自被丢弃的肝脏,因而实验往往不成功”,Camargo在一份新闻稿中说,“这种对细胞来源不加限制的做法,我想现在是时候重新思考是否再利用起来。”

在这项研究中,研究人员在体外培养肝细胞,然后再将它们移植到有基因缺陷的小鼠肝脏中。随着干细胞在未来的几个月里逐渐成熟,小鼠肝脏显示出病变的地方逐渐得到改善。研究者称,未来他们将要探讨肝炎等疾病是如何影响Hippo信号途径来抑制其功能。
