据报道,PTEN第二个最常见人类癌症的基因突变。研究报告发表在《分子细胞》杂志中,研究揭示了PTEN基因损失对人类的影响,以及PTEN基因损失为什么会造成前列腺癌或乳腺癌。这些结果可以帮助识别哪些患者受益于新的靶向治疗。

PTEN称为肿瘤抑制基因,意味着它可以减缓细胞的生长,然而,若其损失就可能会导致癌症。通过调节化学磷脂酰肌醇三羟甲基氨基甲烷磷酸盐的水平,PTEN有助于限制细胞生长,因此也可以预防癌症。

然而,新论文表明这只是癌症的一部分。研究团队已经确定了PTEN可能阻止另一种不受控制细胞生长的方式,PTEN还可以减少另一个类似分子的水平,称为磷脂酰肌醇二磷酸。磷脂酰肌醇二磷酸的作用越来越明显,它可以改变AKT蛋白的活性,这是细胞生长的主要监管机构,也可能影响其他调节入侵过程的蛋白质,影响体内癌细胞扩散和转移。

研究人员表示:“我们对这一结果感到很惊讶,这样的一个损失竟然会引起前列腺小鼠模型磷脂酰肌醇二磷酸的增加,磷脂酰肌醇二磷酸被认为是一个谜,许多人认为它只是磷脂酰肌醇三羟甲基氨基甲烷磷酸盐的一个副产品。我们的工作表明,研究磷脂酰肌醇二磷酸调节过程可以解释为什么PTEN具有强大的肿瘤抑制基因,可以帮助我们确定这一系列癌症的全新治疗目标。
