革兰氏阴性菌具核梭杆菌和牙龈卟啉单胞菌是与牙周疾病相关的复杂牙齿生物膜的成员。

在这项研究中，我们以单物种和双物种生物膜培养了具核梭杆菌和牙龈卟啉单胞菌，并通过高分辨率液相色谱-串联质谱法分析了生物膜细胞外聚合物基质（EPM）的蛋白质组成。无标记的定量蛋白质组学分析用于蛋白质鉴定和基于序列的功能表征，以对其进行分类和预测在EPM中的作用。

结果，我们分别在具核梭杆菌，牙龈卟啉单胞菌和双物种生物膜的基质中鉴定出542、93和280种蛋白质。双物种生物膜中几乎70％的所有EPM蛋白起源于核镰刀菌，其中大多数是胞质蛋白，表明核镰刀菌细胞的裂解作用增强。蛋白质组学分析还表明了这两个物种之间的相互作用：22种具核梭杆菌蛋白显示出单种和双种EPM之间的差异水平，并且在双种EPM中仅检测到11种蛋白（分别来自具核梭杆菌的8种和牙龈卟啉单胞菌的3种）。氧化还原酶和伴侣蛋白是所有三种EPM中鉴定出的最丰富的蛋白质之一。生物膜基质另外还包含几种已知的假想的毒性蛋白，它们可以介导对宿主细胞的粘附和牙周组织的分解。

综上所述，这项研究表明，两种重要的牙周病原体的生物膜基质由数量和功能差异很大的多种蛋白质组成。相对较高水平的几种检测到的蛋白质可能会促进它们潜在地用作抑制生物膜发育的靶标。
