研究人员通过建立Nexn全敲除小鼠和心肌细胞特异性敲除小鼠,进行表型、RNA测序和质谱分析等实验,发现在小鼠中,Nexn全敲除和心肌细胞特异性敲除均可导致一种快速进展的扩张型心肌病。体内外分析表明,NEXN与连接肌浆网蛋白相互作用,对钙的最佳暂态过程至关重要,并参与启动T管内陷和形成。

本研究表明,NEXN是连接膜复合体的关键组成部分,对T管起始和形成必不可少,为解析携带NEXN突变的心肌病患者的潜在致病机制提供见解。
