STM:肾脏损伤不要怕 MG53帮你忙!

近日,美国俄亥俄州立大学的华人科学家Jianjie Ma在国际期刊Science Translational Medicine发表了他们关于MG53保护肾脏细胞修复急性肾损伤的最新研究进展。   MG53蛋白是肌特异性TRIM家族蛋白,是细胞执行自我修复的关键成分。之前研究发现MG53蛋白主要存在于骨骼肌和心肌组织中,能够对心脏,肺和骨骼肌细胞损伤进行修复。在这项研究中,研究人员发现

MG53蛋白是肌特异性TRIM家族蛋白,是细胞执行自我修复的关键成分。之前研究发现MG53蛋白主要存在于骨骼肌和心肌组织中,能够对心脏,肺和骨骼肌细胞损伤进行修复。在这项研究中,研究人员发现MG53对急性肾脏损伤也有重要保护作用。
 
肾脏在许多药物治疗过程中均会受到应激,比如败血症,心内直视手术,化疗等治疗过程,因此急性肾脏损伤是一种比较常见的疾病。如果不能在急性肾损伤早期阶段进行治疗就会演变为慢性肾脏疾病。
 
研究人员将正常肾小管上皮细胞与MG53缺失细胞进行对比后发现,在受到机械损伤后,正常细胞能够存活而MG53缺失细胞会很快死亡。重建MG53表达能够促进MG53缺失细胞的存活,同时没有出现副作用。研究人员将MG53应用于静脉实验发现其能够保护肾脏免受缺血再灌注造成的组织损伤,将MG53与化疗药物cisplatin联合使用也能够起到类似的保护作用,并且不影响化疗药物的肿瘤抑制功能。
 
这项研究表明MG53对于急性肾脏损伤具有重要修复作用,能够保护肾脏免受化疗药物等带来的应激损伤,具有重大应用价值。
