科学家们研究了在怀孕期间暴露于柴油废气颗粒或城市空气颗粒物的孕鼠所生的三代小鼠的健康状况,并将第一代、第二代和第三代小鼠的肺细胞与未接触污染物健康小鼠所生三代进行比较。研究结果表明暴露在柴油废气颗粒或城市空气颗粒物中孕鼠的后代,其一种常见的过敏标志物的免疫细胞异常增加。并且调节免疫系统的白细胞介素水平也明显升高,这表明其后代哮喘患病的风险增加。值得注意的是,第一代和第二代情况更为显着,这表明遗传风险因素也随着代数进一步减少。

出生前暴露与于环境污染会导致子代DNA表观的遗传变化,从而影响DNA甲基化,引起染色质结构、DNA构象、DNA稳定性及DNA与蛋白质相互作用方式的改变,从而控制基因表达。研究人员发现,典型的DNA甲基化所导致的跨代哮喘的风险是由于一种称为树突细胞的免疫细胞发生异常改变。树突状细胞在哮喘早期的发生发展中起关键作用。
