BJC:新策略或可有效降低癌细胞对放疗的耐受性

很多人都知道,利用放疗能够治疗很多类型的癌症,然而有些人却并不会意识到有些癌细胞或许对放疗并没有反应,口腔鳞状细胞癌(OSCC)就是其中一种癌症,近日,来自日本熊本大学的研究人员就通过研究寻找能够抵御癌细胞对放疗产生耐受性的方法,相比直接作用癌细胞而言,他们试图去寻找一种方法来控制细胞的生物学机制从而帮助抵御癌细胞对放疗产生耐受性,相关研究刊登于国际杂志British Journal of Cancer上。

文章中,研究者重点对白细胞介素-6(IL-6)和核转录因子Nrf2进行了研究,白细胞介素-6是一种炎性反应信号因子,而Nrf2则是一种保护机体抵御氧化性应激的关键蛋白;研究者开发的方法包括在两种不同类型的癌细胞样本中进行体外实验,其中一种样品直接来自恶性口腔鳞状细胞癌患者机体的组织,这些患者已经接受了放疗进行治疗(总放疗剂量为30戈瑞);其它样本则包括人类的口腔鳞状细胞癌细胞系,其来自于日本生物资源研究样本库,这些细胞接受的放射剂量为6或10戈瑞。

研究结果发现,IL-6能够通过与Nrf2抗氧化通路相互作用来保护放疗对癌细胞的作用,而且这种相互作用和研究者所发现的来自氧化性损伤的保护作用都非常有意思;本文中研究者首次通过研究发现,IL-6对于Nrf2抗氧化通路具有一定效应,他们希望后期能够以IL-6为靶点来开发新型疗法从而有效对于癌细胞对放疗产生耐受性。

目前大部分都是在体外进行的,而这也是本研究的主要限制,然而研究人员已经对药物托珠单抗(Tocilizumab)免疫抑制的小鼠模型中开展了相关实验,托珠单抗能够用作治疗类风湿性关节炎,其也能够作为一种OSCC的疗法有效抵御IL-6R;未来研究人员将通过更为深入的研究开发新型疗法来有效降低癌细胞对放疗的耐受性。
