使用zeta电位和粒度分布表征（1）GO; （2）GO-PEI; （3）GO-PEI-ASyycG。将重组ASyycG载体转化到MRSA细胞中，测定yycF / G和icaADB基因的表达水平，并通过定量实时PCR（qPCR）测定进行比较。随后用编码增强型绿色荧光蛋白（ASyycG-eGFP）的基因作为报告基因修饰重组ASyycG质粒，并通过荧光强度评估转化效率。通过结晶紫测定，集落形成单位测定和共聚焦激光扫描显微镜评估MRSA菌株的生物膜生物量和细菌活力。

结果表明，GO-PEI-ASyycG的Z-平均尺寸远大于GO或GO-PEI。基于GO-PEI的策略显着提高了ASyycG转化的效率。 GO-PEI-ASyycG转化的MRSA菌株具有生物膜形成相关基因的最低表达水平。此外，GO-PEI-ASyycG在生物膜建立24小时后抑制生物膜聚集并改善对MRSA的杀菌作用。

总之，该研究结果表明，基于GO-PEI的反义yycG RNA将是治疗MRSA感染的有效方法。
