NAT GENET: 睡美人发现新的癌基因

“睡美人”转座子是第一瘤研究中起到了重要作用。通过癌症基因组测序确定的绝大多数遗传改变需要进一步的研究方法来解释其意义和相互作用。

“睡美人”转座子是第一个应用于高等哺乳动物细胞相关研究的转座子，它属于DNA转座子的一种。作为一种简单、有效的转基因工具，近年来，睡美人转座子在肿瘤研究中起到了重要作用。

通过癌症基因组测序确定的绝大多数遗传改变需要进一步的研究方法来解释其意义和相互作用。在这项研究中我们开发了一种新型全身插入诱变筛选小鼠，这一小鼠是为发现Pten协同肿瘤抑制基因而设计的。为了筛选癌基因，我们在同一基因组内耦合失活的单拷贝失活睡美人转座子与失活的Pten。 

这项对278个转座子介导的前列腺，乳腺，皮肤癌的分析显示：一些组织特异性和互相联合的癌基因或候选癌基因与癌症相关。我们检测了ZBTB20，CELF2，PARD3，AKAP13和WAC，经过我们在多种癌症类型中的筛选被鉴定为前列腺癌中的新的肿瘤抑制基因。我们揭示了他们与PTEN在预防体外侵袭的协同作用，并确认其临床相关性。 Wac在体内的进一步研究显示在Pten缺乏条件下，该基因（其编码自噬调节因子）的显示出单倍体不足的表征。我们的研究显示复杂的PTEN合作肿瘤抑制网络在不同的癌症类型，具有潜在的临床意义。
