Sci Adv:3D生物打印新突破:细菌墨水可打印任何三维结构

近年来,科学家一直在开发3D打印技术,随着3D打印制造人类器官的实现,几乎已经没有什么东西不能用它打印出来了。现在,研究人员开发出一种新型3D打印平台,通过使用细菌墨水,可以打印出任何三维结构。

由复合材料实验室的负责人André Studart教授领导的一组苏黎世联邦理工学院(ETH)研究人员开发出一种含有细菌的墨水,并将它命名为“Flink”,即“功能活性墨水”。研究成果发表在近期《Science Advances》杂志上。

墨水是由一种水凝胶和细菌混合构成。水凝胶本身是由透明质酸、长链糖分子和热解硅石构成的。细菌的培养基混合到墨水中,使其具有细菌生存的所有先决条件。使用这种凝胶作为基础,研究人员可以添加具有所需的“性能”的细菌,然后打印出他们喜欢的任何三维结构。

第一作者、Studart的小组成员Patrick Ruhs和Manuel Schaffne在他们的工作中使用了恶臭假单胞菌(putida)和木醋杆菌(Acetobacter xylinum)。前者是可分解化学工业大规模生产的有毒化学苯酚,后者则可以分泌高纯度的纳米纤维素。这种细菌纤维素可以缓解疼痛,保持水分,稳定性强,从而开辟了在烧伤治疗中的潜在的应用。

像牙膏一样粘稠

在开发墨水的过程中,水凝胶的流动特性提出了一个特殊的挑战:墨水必须足够流动,才能通过压力喷嘴。墨水的一致性也影响细菌的流动性,墨水越硬,细菌越难移动。然而,如果水凝胶太硬,则醋杆菌分泌的纤维素较少。与此同时,打印品必须足够坚固,以支撑后续层的重量。如它们太流体,就不可能打印出足够稳定的结构,因为它们会在重压之下坍塌。

Schaffner 总结墨水的关键,“墨水必须像牙膏一样粘稠,并具有护手霜的一致性。”

到目前为止,材料科学家还没有研究过印刷品里细菌的寿命。Ruhs说,“由于细菌对资源的需求非常少,所以我们认为它们能够在印刷品中存活很长一段时间。”

巨大的应用潜力

目前,这项研究仍处于初级阶段,Ruhs说,“使用细菌墨水打印具有巨大的潜力,大多数人只把细菌与疾病联系在一起,但是实际上如果没有细菌,我们是无法生存的。”研究人员相信他们的细菌墨水是完全安全的,因为使用的细菌都是无害且有益的。

除了医学和生物技术的应用,研究人员还设想了许多其他的潜在用途。例如,这种物质可用于研究降解过程或生物膜形成。一个实际应用可能是含有细菌的3D打印传感器,用于检测饮用水中的毒素。另一个想法是制造含有细菌的过滤器,用于灾难性的石油泄漏。

想要实现这些应用,首先需要克服3D生物打印时间慢、扩展性差的挑战。目前,木醋杆菌需要数天的时间才能产生用于生物医学应用的纤维素。科学家们相信,他们可以进一步优化和加速这个过程。
