囊性纤维化肺疾病（CF）是由与上皮钠通道（ENaC）结合的囊性纤维化跨膜传导调节子（CFTR）功能缺失所造成的。在肺脏中，ENaC是负责钠离子运动，其创建了一个渗透压梯度，从而将液体从气道中排出。过度活化的ENaC有助于降低气道水合作用导致粘液脱水，粘液纤毛清除功能降低，慢性细菌感染。ENaC是所有CF患者独立于CFTR突变的潜在治疗靶点。

近日，呼吸内科以及重症医学领域权威杂志Am J Resp Crit Care上发表了一篇研究文章，研究人员旨在评估自然调节功能为控制ENaC活性的SPLUNC1模拟物肽SPX-101在体外和体内实验中的疗效。

研究人员表面生物素化和随后的免疫印迹分析在来自于健康人和CF患者的原代人类支气管上皮细胞上评估了SPX-101的内化。SPX-101的体内治疗效果是在存活的βENaC转基因小鼠中进行评估的，以及这些小鼠的粘液运输和CF绵羊模型的粘液运输。

SPX-101选择性地结合ENaC，并且可以促进的α、β和γ亚基的内化。SPX-101可以使ENaC从膜上移除引起阿米洛利敏感的电流显著降低。SPX-101每日一次的剂量可以使超过90%的βENaC转基因小鼠生存延长。SPX-101可以增加βENaC小鼠模型以及CF绵羊模型的粘液运输。

这些数据表明SPX-101促进ENaC膜浓度持续减低，导致粘液运输显著改善。
