之前,研究人员表明了突变的FGFR3能够改变上皮细胞的细胞与细胞间和细胞与基质间的连接特性,从而导致接触抑制的扩散。最近,有研究人员调查了ETS家族转录因子的ETV5,即一个假定的膀胱癌中FGFR3信号的效应因子。研究发现,FGFR3信号能够诱导MAPK/ERK调控的ETV5水平的增加,并且最终导致TAZ水平的增加;TAZ是Hippo信号途径下游的转录共调控因子,且与细胞接触抑制有关。研究人员还阐释了ETV5是突变FGFR3致瘤效应的一个关键下游调节因子,并且在FGFR3突变膀胱细胞系中ETV5的敲除与增殖的减少和不依赖贴壁生长的减弱有关。

最后,研究人员指出,他们的结果促进了人们对上皮恶性肿瘤转化过程中发生的分子变异的理解,并表明了TAZ在FGFR3依赖的膀胱肿瘤中可能作为治疗靶标潜力。
