PNAS:“化疗明星”紫杉醇导致癌转移,要怪这个基因

紫杉醇(Paclitaxel,PTX)别名红豆杉醇,泰素,紫素,特素,是目前已发现的最优秀的天然抗癌药物,在临床上已经广泛用于乳腺癌、卵巢癌和部分头颈癌和肺癌的治疗,被称作“化疗明星”。

紫杉醇促进癌转移,与Atf3基因有关

然而最新研究发现:紫杉醇是把双刃剑,在利用细胞毒性杀死癌细胞的同时,也会增加癌细胞对化疗剂的抗性并促进癌细胞转移,机制跟Atf3基因密切相关。文章发表在近期出版的《美国国家科学院院刊》(PNAS)上。

来自美国俄亥俄州立大学生物化学与药理学教授 Tsonwin Hai 团队的一项研究为紫杉醇促进乳腺癌的肺转移提供了“佐证”。他们对能够产生自发转移的乳腺癌模型小鼠注射了一定量的紫杉醇,结果发现原发肿瘤确实有缩小,但转移到肺部的癌细胞却更多。于是,研究者将一部分小鼠的Atf3 基因进行敲除,结果发现转移到肺部的癌细胞明显减少。

上述图片指的是:在第26天,不使用紫杉醇和使用紫杉醇及敲除Atf3基因后不使用紫杉醇和使用紫杉醇的四种小鼠肺转移肿瘤占总肺部的面积,结果说明紫杉醇的这种促进癌症转移的作用与Atf3基因有关,对这几种肿瘤组织进行了检测,发现用紫杉醇治疗的小鼠,与血管生成有关的一些基因表达量高,与抗血管生成有关的一些基因表达量低。

改变肿瘤转移微环境



紫杉醇对肿瘤织的影响

肿瘤转移微环境由三类交织在一起的不同细胞组成:表达MENA蛋白的肿瘤细胞,血管周围的巨噬细胞以及内皮细胞。这三种细胞的密度和乳腺癌转移的风险挂钩。

在这项研究中,他们还发现,使用紫杉醇治疗的小鼠肿瘤转移微环境密度明显升高,而Atf3基因介导的化疗剂对癌症肺转移的促进作用是化疗中普遍存在的现象。

无独有偶,几个月前知名科研机构爱因斯坦医学院(Albert Einstein College of Medicine)的研究人员发表在《科学》子刊《Science Translational Medicine》上的一项研究也揭示:在往化疗的药物中添加紫杉醇后,小鼠体内的“肿瘤转移微环境”有所增多,患者的完全缓解率有着显著上升,但却不能延长患者的总体生存率。

抑制Atf3基因表达,解除化疗副作用

综上所述,紫杉醇不仅会改变原发肿瘤周边血管的状态,帮助乳腺癌细胞进入血循环;还会改造肺组织微环境,使更适合癌细胞生存;而导致这两件事发生的主要原因均与Atf3基因有关。因此,通过抑制Atf3基因的表达,有望解除化疗的副作用,这也是 Tsonwin Hai 团队的下一个研究目标。
