Clin oral invest:重组人骨形态发生蛋白-2和-9对大鼠颅骨临界缺损后骨形成影响的比较

在骨形态发生蛋白(BMP)家族成员中,BMP-2和BMP-9已经显示出有力的骨诱导潜能。然而,体内骨再生潜力的差异仍不清楚。本研究旨在比较重组人(rh)BMP-2和rhBMP-9对大鼠颅骨临界缺损(CSD)骨形成的影响。

在骨形态发生蛋白(BMP)家族成员中,BMP-2和BMP-9已经显示出有力的骨诱导潜能。然而,体内骨再生潜力的差异仍不清楚。本研究旨在比较重组人(rh)BMP-2和rhBMP-9对大鼠颅骨临界缺损(CSD)骨形成的影响。

材料和方法:28只Wistar大鼠在每个顶骨两侧制备两个颅骨缺损。缺损(n = 56)分为4组:吸收性胶原海绵(ACS),rhBMP-2+ACS(rhBMP-2 / ACS),rhBMP-9 / ACS或假手术(对照) rhBMP-2 / ACS和rhBMP-9 / ACS的治疗或相同的治疗不包括在相同的动物中。术后2周和8周处死动物。通过微型计算机断层扫描分析颅骨缺损的面积(BV),组织学分析缺损闭合百分比(DC / DL),新形成的骨面积(NBA / TA),骨髓面积(BMA / NBA),脂肪组织面积(ATA / NBA),中心骨高度(CBH)和边缘骨高度(MBH)。

结果:rhBMP-2 / ACS组术后BV(5.44±3.65 mm3,n = 7)大于术后2周,术后8周时与对照组和ACS组相比(6.02±2.90和9.30±2.75 mm3,n = 7),rhBMP-2 / ACS组和rhBMP-9 / ACS组(分别为18.17±2.51和16.30±2.46 mm3,n = 7)BV值更高。与对照组和ACS组相比,rhBMP-2 / ACS和rhBMP-9 / ACS组在术后8周诱导新骨形成明显。然而,在任何组织形态学分析中rhBMP-2 / ACS和rhBMP-9 / ACS组之间没有显着的统计学差异。 rhBMP-2 / ACS组的ATA / NBA(9.24±3.72%,n = 7)在术后8周实验组最高。

结论:在本研究的范围内,可以推断出rhBMP-2 / ACS在2周时引起新骨形成的早期轻微增加,在大鼠CSD的8周愈合期后的组织,与rhBMP-2 / ACS相比,rhBMP-9 / ACS形成的新骨具有较少脂肪。
