基质金属蛋白酶(MMP)是一类依赖锌离子切割多肽的广谱性蛋白酶。此前的研究发现巨噬细胞的激活能够引发MMP-25表达量的上调,体外的功能性试验也证明MMP能够有效切割胞外基质的成分,比如IV型胶原。相关研究表明MMP-25参与了多种疾病的发生,其中包括多发性硬化,但其具体的功能却不得而知。为了研究这一问题,来自西班牙奥维耶多大学的Carlos Lopez-Otın课题组进行了深入研究,相关结果发表在最近一期的《Journal of Immunology》杂志上。

首先,作者构建了MMP-25缺失突变型小鼠,并验证了其MMP-25的表达。进一步,作者通过血液检测发现突变小鼠患有高丙种球蛋白血症,而且其体内的IL-1a的表达量相应较低。

为了研究MMP-25在天然免疫中的作用,作者向野生型小鼠与突变体小鼠分别注入了LPS进行刺激,结果显示,突变体小鼠受到LPS刺激后的存活率明显高于对照组。另外,突变体小鼠肺部的炎症损伤程度较对照组也明显降低。这说明MMP-25对于炎症反应的发生具有正向的调控作用。

进一步,作者发现突变体小鼠的巨噬细胞在受到LPS刺激后NF-KB的激活受到了抑制。另一方面,作者通过体外实验发现MMP-25能够促进TRAF6的泛素化。以上实验证明MMP-25能够促进TRAF6的泛素化,从而介导LPS引起的NF-KB的激活以及炎症反应。
