Oncogene:UHRF1能够通过对TXNIP的表观调控促进肾细胞癌的恶化

最近,有研究人员报道了RCC肿瘤组织比正常肾组织具有明显更高的UHRF1表达。siRNA或者shRNA引起的UHRF1表达的下调在RCC细胞系中能够导致细胞生存力减少、抑制细胞迁移和侵入,以及增加细胞凋亡。UHRF1的敲除RCC异种种植模型同样能够导致明显的体内肿瘤生长的抑制。在RCC细胞中下调UHRF1表达后,TXNIP的表达上调。另外,与UHRF1单独下调细胞系相比,当UHRF1和TXNIP同时下调后,细胞生存和细胞侵入能力增加,而细胞凋亡减少。研究人员还展示了UHRF1能够招募HDAC1到TXNIP启动子,调控组蛋白H3K9的去乙酰化,导致TXNIP表达的抑制。

最后,研究人员指出,他们的结果确定了UHRF1在RCC中具有致瘤功能,并且UFRF1也许能够通过对TXNIP的表观调控来促进肿瘤恶化。UHRF1也许可以作为RCC治疗的一个靶标。
