Sci Rep最新的一篇研究报告,研究者评估了9-oxo-ODAs对人宫颈癌细胞的抗肿瘤作用。在体外证实了9-oxo-ODAs的抗肿瘤作用,通过组学分析确定了它们的作用机制,并在体内外进行了验证。

9-oxo-ODAs呈浓度依赖性抑制人宫颈癌细胞系(HeLa和SiHa)的细胞增殖(IC50= 25 ~ 50µM)。FCM分析显示9-oxo-ODAs诱导细胞凋亡。转录组学、蛋白质组学和富集分析表明,9-oxo-ODAs治疗显著改变了细胞周期和p53通路,并降低了细胞周期蛋白依赖性激酶1 (CDK1)蛋白表达。实时定量PCR结果显示,9-oxo-ODAs呈浓度依赖性地降低CDK1 mRNA的表达。在体外实验中,9-oxoODAs降低了HPV癌蛋白的表达。在离体人宫颈癌组织中,9-oxo-ODAs降低了CDK1的表达,增加了凋亡标志物cleaved caspase 3的表达。此外,9-oxo-ODAs在体内显示出抑制宫颈癌转移和生长的潜力。

综上,9-oxo-ODAs诱导hpv阳性人宫颈癌细胞的细胞周期阻滞和凋亡,这一过程涉及CDK1。因此,9-oxo-ODAs可能是宫颈癌的潜在治疗药物。
