几十年来,科学家一直在试图利用癌细胞这种贪婪的新陈代谢,作为抗癌新疗法的靶标。最近,美国杜克大学的研究人员,发现了肾细胞癌的一个有用靶标。2016年2月1日他们在Cancer Research发表的一项研究表明,这些癌症能够以一种方式改变代谢,这种方式使癌细胞对外界的一种营养物(称为胱氨酸)成瘾。通过剥夺癌细胞的胱氨酸,研究人员能够在这种疾病的小鼠模型中,触发一种形式的细胞死亡,称为细胞坏死。

本文资深研究作者、杜克大学医学院分子生物学和微生物学副教授Jen-Tsan Ashley Chi指出:“我们发现,使这些肿瘤如此具有侵袭性的同一机制,也使其更容易出现营养缺乏。这就像是,我们用它自己的方式击败它们。”

Chi表示,肾细胞癌——一种形式的肾癌,历来是很难治愈的,这项研究指出了一种有前途的新方法。这种疾病每年夺去100000多人的生命。大约四分之三的肾细胞癌病例是由一个缺失的VHL肿瘤抑制基因所标记,这个基因可使健康的细胞发展成肿瘤。Chi和本文第一作者、实验室博士后Xiaohu Tang,决定探究这个遗传变化如何可能影响癌细胞的代谢和营养需求。

Tang使癌细胞经受了营养剥夺试验,一个接一个地从其生长介质中除去了15种氨基酸。大部分的时间,细胞能够相当好地经受住这些变化,减缓它们的生长,但仍然保持健康。但是Tang发现,当胱氨酸被去除之后,细胞肿胀并浮到表面,这是坏死性死亡的一个迹象。

研究人员随后进行了一系列的遗传分析,将负责这一营养成瘾的基因网络拼凑起来。通常,VHL基因可抑制另一个基因——被称为肿瘤坏死因子α(TNF-α)。当VHL缺失时,TNF-α的高水平可致使负载有危险自由基的癌症更快的生长、更具有侵袭性。

胱氨酸负责维持高水平的抗氧化剂——消除氧自由基;所以,当研究人员去除了这种营养物时,癌细胞基本上死于它们自己的“自由基损伤”之手。

研究人员表明,无论在组织培养细胞还是小鼠模型中,该方法都获得了成功。Tang和杜克大学癌症研究所David Hsu实验室的同事们一起,将肾细胞癌肿瘤移植到小鼠中,然后用柳氮磺胺吡啶——一种阻断胱氨酸的药物,来治疗动物。他们发现,治疗可引起细胞坏死,并明显地延迟了肿瘤生长。

阻断糖酵解 饿死癌细胞
