9月13日,发表在Scientific Reports上的一项研究中,英属哥伦比亚大学(University of British Columbia,UBC)的科学家们发现了癌细胞是如何躲过免疫系统监视的。这是肿瘤转移和扩散至全身的关键一步。

该研究的通讯作者Wilfred Jefferies教授说:“免疫系统能够有效识别和阻止原发性肿瘤的发生和扩散,但当转移性肿瘤出现时,免疫系统就不再能识别癌细胞并阻止它们了。

这一研究发现了一种新的、能够解释转移性肿瘤如何战胜免疫系统的机制。同时,科学家们发现,逆转这一机制能够让肿瘤“脱下隐身衣”。

具体来说,研究人员发现,随着癌细胞的发展,它们可能失去了产生interleukein-33(IL-33,一种蛋白质)的能力。当IL-33从肿瘤中消失时,免疫系统就无法识别癌细胞了。

IL-33如何发挥作用?

事实上,IL-33会影响另一种蛋白复合体——主要组织相容性复合体(MHC)。当这些蛋白正常工作时,原发性肿瘤细胞外会出现“警告标志(warning flags)”,这样免疫细胞就能识别并摧毁它。但当肿瘤中IL-33消失后,这种标志展示通路(flag-displaying pathway)会瓦解,癌细胞就能躲过免疫系统的“追捕”,开始扩散或转移。

该研究对数百名病人进行了调查,结果发现,前列腺癌或肾癌患者的肿瘤丢失了IL-33,五年内癌症复发更快。现在,研究人员开始调查,是否检测IL-33是监控某些类型癌症进展的有效途径。论文的第一作者Iryna Saranchova说:“IL-33可能成为前列腺癌的首批免疫生物标志物。”

很长时间以来,研究人员都在尝试利用免疫系统来对抗癌症。在过去的几年里,这种思路终于展现了它的潜能。这一研究证实,将IL-33“放回”转移性癌症中能够帮助恢复免疫系统识别肿瘤的能力。进一步的研究将调查这一方法能否成有效的抗癌疗法。
